You must
A: Be of good moral character (no violations, stay on the right side of the law, courteous)
B: Be knowledgeable on the relevant field (guides by you, etc)
C: Help out a lot in general
D: Be around when a new board comes along (Next is probably EW5 or WC4)
E: Be picked by the existing moderators
I know because...I've been through it

Manstein became a moderator before he even had 100 posts, so post count doesn't affect who can be a moderator.
